چکیده

A B S T R A C T Five patients with glucagonomas had elevated plasma levels of total glucagon immunoreactivity. Gel filtrations of these plasma samples on Bio-Gel P30 columns showed that most of the immunoreactivity eluted in the 3,500-(true glucagon) and 9,000-dalton fractions. After the administration of alpha cell effectors, changes in total glucagon immunoreactivity were seen which were accounted for primarily by the 3,500-dalton species, but there were also changes in the 9,000-dalton moiety. Venous effluent plasma from tumors of two subjects contained elevated concentrations of glucagon immunoreactivity in both fractions. When material from both the 3,500and 9,000-dalton peaks were serially diluted in a glucagon immunoassay, parallel displacement curves were found, suggesting that both have similar or identical antigenic determinants. Thus, with conversion to a neoplastic state, alpha cells of glucagonomas, much like beta cells of insulinomas, may secrete an increased amount of a larger, 9,000mol wt glucagon species which may be a prohormone.

Effect of esophageal distension on gastric secretions in rat and the possible mechanism

Introduction: The effect of esophageal distension (ED) on gastric motility has been well documented, but a few investigations have been carried out on the effect of ED on gastric secretion. The aim of this study was to investigate the effect of ED on gastric acid and pepsin secretion and the mechanism(s) involved. Methods: Male adult Wistar rats (200-240 g) were anesthetized with urethane (1...

Management of the hormonal syndrome of neuroendocrine tumors

Gastroenteropancreatic neuroendocrine tumors (GEP/NET) are unusual and rare neoplasms that present many clinical challenges. They characteristically synthesize store and secrete a variety of peptides and neuroamines which can lead to the development of distinct clinical syndrome, however many are clinically silent until late presentation with mass effects. Management strategies include surgery ...

Effect of thyroid hormones on basal and stimulated gastric acid secretion due to histamine, carbachol, and pentagastrin in rat (an in vitro study)

Although thyroid hormones are known to influence acid secretion, but exact mechanisms are not fully understood. So, in this experimental study carbachol, histamine, and pentagastrin were used to stimulate acid secretion in isolated stomach of hypo- and hyperthyroid rats and data were compared with controls. Each group consisted of 8 N-Mari rats from both sexes weighing 246 ± 5 g. Hypo- and hype...

Do glucagonomas always produce glucagon?

Pancreatic islet α-cell tumours that overexpress proglucagon are typically associated with the glucagonoma syndrome, a rare disease entity characterised by necrolytic migratory erythema, impaired glucose tolerance, thromboembolic complications and psychiatric disturbances. Paraneoplastic phenomena associated with enteric overexpression of proglucagon-derived peptides are less well recognized an...
